Sejnowski Computational Neurobiology Lab, Salk Institute, La Jolla, CA, USA The low-threshold Ca2+ channels that mediate regenerative Ca2+ currents in central neurons are responsible for generating bursts of Na+/K+ action potentials. In this modeling study we explored kinetic parameters of T-channels for their impact on burst latency and strength. The figure below shows a tradeoff occurring between latency and strength across the ranges of time constants for 1G, H, and I subtypes. We propose alternative assays of bursting that may bear directly on the organization of the encoding gene, with implications for the evolution of these channels. Supported by: NIH HHMI

T-type calcium channels in burst-firing, network synchrony, and epilepsy.

Low voltage-activated (LVA) T-type calcium channels are well regarded as a key mechanism underlying the generation of neuronal burst-firing. Their low threshold for activation combined with a rapid and transient calcium conductance generates low-threshold calcium potentials (LTCPs), upon the crest of which high frequency action potentials fire for a brief period. Experiments using simultaneous ...

Functional impact of alternative splicing of human T-type Cav3.3 calcium channels.

Low-voltage-activated T-type (Cav3) Ca2+ channels produce low-threshold spikes that trigger burst firing in many neurons. The CACNA1I gene encodes the Cav3.3 isoform, which activates and inactivates much more slowly than the other Cav3 channels. These distinctive kinetic features, along with its brain-region-specific expression, suggest that Cav3.3 channels endow neurons with the ability to gen...

Contributions of T-type calcium channel isoforms to neuronal firing.

Low voltage-activated (LVA) T-type calcium channels play critical roles in the excitability of many cell types and are a focus of research aimed both at understanding the physiological basis of calcium channel-dependent signaling and the underlying pathophysiology associated with hyperexcitability disorders such as epilepsy.  These channels play a critical role towards neuronal firing in both c...

Pharmacology of sodium and calcium channel modulation in neurons: implications for neuroprotection.

It is now clear that the wide variety of neuronal voltage-dependent calcium channels (Table 1) are determinants of transmitter release (N-type, P-type, Q-type channels), repetitive or burst neuronal firing (T-type channels) and perhaps of some aspects of gene expression (L-type channels). However, although the calcium overload hypothesis of neuronal cell death has been around for 14 years, ther...

Dendritic control of spontaneous bursting in cerebellar Purkinje cells.

We investigated the mechanisms that contribute to spontaneous regular bursting in adult Purkinje neurons in acutely prepared cerebellar slices. Bursts consisted of 3-20 spikes and showed a stereotypic waveform. Each burst developed with an increase in firing rate and was terminated by a more rapid increase in firing rate and a decrease in spike height. Whole-cell current-clamp recordings showed...
